CIALIS

CIALIS (tadalafil) is an oral drug for the treatment of Erectile Dysfunction, belonging to PDE-5 inhibitors class of medications (VIAGRA, LEVITRA).

Tablets and Dosage

Yellow film-coated Cialis (tadalafil) tablets are supplied in 2.5 mg, 5 mg, 10 mg or 20 mg dosage.

Indications

CIALIS is indicated for the treatment of Erectile Dysfunction in males 18 years old. Taken on ‘as needed’ basis.

Dosing

Adults: Each tablet is taken as needed. The initial dose is 10 mg, taken at least 30 min. before sexual activity. The next intake cannot be earlier than after 24 hours. The dose may be increased to 20 mg.

In adults with impaired renal and hepatic functions 10 mg is the maximum dosage.

Long time daily use of doses of 10-20 mg is not recommended.
2.5-5 mg daily dosages of tadalafil are recommended for continuous treatment.

Contraindications

  1. Concomitant therapy with nitrate preparations.
  2. Unstable angina, severe heart failure, hypotension, and recent history of myocardial infarction (3 months) or stroke (6 months).
  3. Tadalafil is contraindicated in patients suffering from non-arteritic anterior ischemic optic neuropathy (NAION), or who have had blood clots in the eye.

Cautions

Cautions should be exercised in patients with:

  1. Anatomical defects of the penis.
  2. Severe hepatic and renal insufficiency.
  3. Serious cardiovascular events, such as acute myocardial infarction that have been reported in connection with use of tadalafil, but definite causal relationship has not been shown.
  4. with hypotension during sexual intercourse.

Tadalafil should not be taken in case of concomitant treatment with α-blockers.
In case of sudden visual defect, the patient should stop taking tablets and see the doctor immediately.
CIALIS can cause dizziness and affect the ability to drive or operate hazardous machinery.

Side effects

  1. Very common (more than 10%): Dyspepsia. Headache.
  2. Common (1-10%): Abdominal pain, gastro-oesophageal reflux. Nasal congestion, facial flushing. Back pain, myalgia. Dizziness.
  3. Uncommon (0,1-1%): Eye pain.
  4. Very rare (less than 0.01%): Serious cardiovascular events, hypotension.

By use of PDE5 inhibitors a number of cases of sudden decrease or loss of hearing, and few cases of visual field defects, vascular occlusion in the retina and non-arteritic anterior ischemic optic neuropathy (NAION) have been reported. In case these side effects occur, contact your doctor immediately.

Interactions

  1. Concomitant treatment with nitrates can cause severe hypotension.
  2. Risk of developing postural hypotension during concomitant treatment with α-blocking agents.
  3. Ketoconazole inhibits the metabolism of tadalafil.
  4. Interactions may also be expected for other drugs metabolised by CYP3A4 enzyme such as erythromycin, itraconazole and HIV protease inhibitors.
  5. Rifampicin increases the metabolism of tadalafil.

Pregnancy / Breastfeeding

Not applicable.

How it Works?

Vardenafil selectively inhibits the breakdown of cyclic guanosine monophosphate (cGMP) in the corpus cavernosum by inhibiting PDE5 enzyme. This restores the natural erectile response. Sexual stimulation is necessary for tadalafil to be effective. Duration of effects is up to 36 hours.
